建立在蛋白质变性-复性三态模型的基础上, 给出了一个描述在变性液中变性蛋白质复性时蛋白质浓度和其复性率的关系式. 通过这个关系式, 可以获得两个重要的描述蛋白质变性-复性体系特征的参数, 一个是包含在一个集聚体分子中的变性蛋白质的分子数目n, 另一个是蛋白质从原始态到形成集聚体过程中的表观集聚平衡常数K. 以三种溶菌酶在脲和盐酸胍溶液中的变性-复性过程对此方程进行了验证, 结果表明所给出的方程能够很好地描述三种溶菌酶在这两种变性液中的复性结果, 三种溶菌酶在两种变性液中有形成二分子集聚体的趋势. 变性溶菌酶在复性过程中的电泳和高效凝胶排阻色谱也同时能够监测到复性过程中集聚体的形成, 并且监测结果与上述方程所得的结果一致. 相似文献

We study the kinetic behaviour of the growth of aggregates driven by reversible migration between any two aggregates. For a simple model with the migration rate K(i;j)=K′(i;j)∝i^uj^v at which the monomers migrate from the aggregates of size i to those of size j, we find that the aggregate size distribution in the system with u+v≤3 and u<2 approaches a conventional scaling form, which reduces to the Smoluchovski form in the u=1 case. On the other hand, for the system with u<2, the average aggregate size S(t) grows exponentially in the u+v=3 case and as (tlnt)^{1/(5-2u)} in another special case of v=u-2. 应用稳态和动态荧光光谱方法,包括荧光探针、标记荧光和荧光淬灭等研究了共聚物P(AM/POEA)在水溶液中的自缔合性质.这种共聚物由丙烯酰胺(AM)和少量疏水单体2-苯氧乙基丙烯酸酯(POEA)组成.实验结果表明这类共聚物的自缔合行为和聚集体结构主要取决于聚合物的链结构和浓度.由胶束共聚合方法得到具有多嵌段结构的共聚物,它们容易发生疏水缔合,并产生很强的增粘作用,而由普通共聚合方法得到的无规共聚物却没有这些性质.同时这类聚合物的缔合类型及其增粘能力也直接与共聚物中的疏水体含量相关,随疏水体含量增加,由于分子间和分子内缔合的竞争,出现粘度先增后降的现象.在荧光研究的基础上提出了多分子聚合物聚集体的结构模型,它随聚集体浓度增加,进一步形成多联聚集体和网状结构.同时还观察到聚集体中疏水体有序排列现象. 相似文献

采用大分子单体法制备接枝型两亲共聚物, 通过表面张力仪、偏光显微镜、旋转流变仪和小角X射线衍射研究了两亲共聚物在水溶液中的聚集行为及其相结构. 首先末端带有可聚合双键的聚甲基丙烯酸叔丁酯大分子单体(PtBMA Macromonomer)通过催化链转移聚合法制备, 所用到的催化链转移剂为二水合双(二氟化硼苯二酮肟)合钴(II) (COPhBF). 然后将所得到的大分子单体与丙烯酸正丁酯(BA)进行自由基共聚得到接枝共聚物PBA-g-PtBMA, PBA-g-PtBMA中PtBMA的侧链部分在酸性条件下定量水解成聚甲基丙烯酸(PMAA)并用NaOH中和得到主链疏水侧链亲水的接枝型两亲共聚物PBA-g-P(MAA-Na+). 用Wilhelmy吊片法研究了不同浓度的两亲共聚物水溶液的表面张力, 发现其行为与小分子表面活性剂不同. 同时用动态光散射法测量了两亲共聚物水溶液中聚集体的粒度, 发现在研究的浓度范围内(0.02~10 g/L)聚集体都存在两个粒度分布峰(约30和300 nm). 浓溶液(w=37.5%)的偏光显微照片呈现层状液晶的特征图案, 而流变研究表明此时体系具有明显的粘弹性, 说明体系形成了层状液晶. 并且用小角度X射线衍射测定了层状液晶的层间距, 约为12.6 nm. 相似文献

Soil aggregate stability as a key indicator of soil structure, is a product of interactions between soil environment, management practices, and land use patterns. The objective of this study was to analyze the impact of various land use patterns on soil aggregate stability in Sichuan Basin of southwestern China. The dry- and water-stable aggregate size distributions were determined by manual dry sieving procedure and Yoder's wet sieving procedure, respectively, while microaggregates and its mechanical and chemical stabilities by Kachisky's method, oscillator method, and citrate-dithionate (C-D) reagent method, separately. The results indicated that fractal dimension and surface fractal dimension were useful indicators to reflect soil aggregate distribution. Land use patterns have an obvious influence on soil aggregate stability. In the study area, water stability, mechanical stability, and chemical stability followed the sequence, Barren landforestlandorchardcropland, and the original stability and collapse velocity were sensitive to soil properties and soil structure. The difference of aggregate stability under different land use patterns is mainly due to the intensity of human disturbance and cultivation. Improper land use patterns will lead to breakdown of unstable aggregates, producing finer and more-easily transportable particles and microaggregates. In the future, inappropriate cultivation and land use patterns should be changed to protect soil structure, to improve soil aggregate stability and soil fertility in Sichuan Basin. 相似文献

Formation of micelle‐type assembly from supramolecular complexation of a surfactant and an oppositely charged homopolymer is demonstrated. The lower CAC observed for these assemblies suggest that the electrostatic interaction provides an amphiphilic homopolymer‐like structure. Spontaneous stereocomplex aggregation of diblock poly(styrene)‐b‐poly(L ‐lactide) PS‐b‐PLLA/poly(D ‐lactide) PDLA pairs has been investigated under ambient temperature in tetrahydrofuran solution. First, diblock PS260‐b‐PLLA165 and PS260‐b‐PDLA162 bearing similar lengths of respective PLLA and PDLA blocks were synthesized through controlled atom‐transfer radical polymerization of styrene, and a subsequent living ring‐opening polymerization of optically pure lactides, and their structures were further characterized by nuclear magnetic resonance spectroscopy (NMR) and gel‐permeation chromatography (GPC). Subsequently, new enantiomeric poly(D ‐lactide) stabilized core‐shell fluorescent CdSe quantum dots (CdSe/PDLA QD) were designed and prepared as sensitive fluorescence labels to shed new lights on the spontaneous stereocomplex aggregation in THF, which was mediated by stereocomplexation of the PLLA and PDLA chains. Upon simply mixing two individual THF solution of diblock PS260‐b‐PLLA165 and HO‐PDLA30‐SH, spontaneous stereocomplex aggregation was studied, and the aggregated uniform spherical particles were observed by scanning electronic microscopy (SEM) to exhibit average particle diameters of 2.0 μm. Finally, utilizing the prepared CdSe/PDLA QDs as new fluorescent labels, morphologies of the spontaneous aggregates by new diblock PS260‐b‐PLLA165/HO‐PDLA30‐SH pair were for the first time directly visualized by a confocal laser scanning fluorescence microscopy (CLSFM). These results might suggest alternative ways to simply prepare functional fluorescent particles with tunable diameter sizes and would be helpful to understand the mechanism of stereocomplex particle aggregation. © 2009 Wiley Periodicals, Inc.
